Patch clamp techniques for single channel and whole-cell recording

چکیده
The patch clamp technique was first used by Neher and Sakmann (1976) to resolve currents through single acetylcholine-activated channels in cell-attached patches of membrane of frog skeletal muscle. The method they used (described by Neher, Sakmann & Steinbach, 1978) and subsequent refinements (Hamill, Marty, Neher, Sakmann & Sigworth, 1981) have led to techniques for high resolution recording of current in excised membrane patches in addition to those that remain cell-attached. Single channel recording yields information about unitary conductance and kinetic behaviour of ionic channels already partly investigated by classical voltage clamping and by noise analysis; it is also leading to the discovery of new classes of ion channel. Together with the method of whole-cell recording, which permits the application of voltage clamping to cells that are too small for microelectrode methods, patch clamp techniques also permit investigations of the physiological role of ionic channels in cells otherwise inaccessible to voltage clamp and to cells that are not electrically excitable.

Development of Automated Patch Clamp Technique to Investigate CFTR Chloride Channel Function
The chloride (Cl-) channel cystic fibrosis transmembrane conductance regulator (CFTR) is defective in cystic fibrosis (CF), and mutation of its encoding gene leads to various defects such as retention of the misfolded protein in the endoplasmic reticulum, reduced stability at the plasma membrane, abnormal channel gating with low open probability, and thermal instability, which leads to inactiva...

Present whole-cell patch-clamp methodology has only moderate consistency and throughput, rendering impractical functional measurements on large numbers of ion channel ligands or on large numbers of unknown or mutant channel genes. In the population patch clamp (PPC) described herein, a single voltage-clamp amplifier sums the whole-cell currents from multiple cells at once, each sealed to a sepa...

1. Since the inception of the patch-clamp technique, single-channel recording has made an enormous impact on our understanding of ion channel function and its role in membrane transport and cell physiology. 2. However, the impact of single-channel recording methods on our understanding of intracellular Ca2+ regulation by internal stores is not as broadly recognized. There are several possible r...

The combination of patch-clamp and molecular biology techniques has made it possible to characterize the pharmacological and biophysical properties of ion channels in single neurons and to screen for expression of specific mRNAs in the same cell. Following whole-cell recording, the cytoplasm of the cell is harvested, and RNA is reverse transcribed into cDNA and amplified in PCR with primers spe...
متن کاملWhole-Cell Patch-Clamp Recordings
The patch-clamp recording technique, which measures ionic currents under voltage clamp, was designed to study small patches of membranes in which near perfect control of the transmembrane voltage can be achieved readily. The recent application of patch-clamp methodology to the analysis of whole-cell current actually defies many of the original design requirements. Nevertheless, whole-cell recor...
